An active research program since 2016

Bazelet has run an active research and development program since 2016, and its science team is among the most productive in the field. We design standardized, terpene-enriched formulations to defined cannabinoid and terpene compositions, built on the interactions between cannabinoids and terpenes, and we test them through preclinical studies and randomized, double-blind, placebo-controlled clinical trials with leading Israeli medical centers. Our work is published in peer-reviewed journals, including JAMA Surgery, Biochemical Pharmacology, Molecules, Frontiers in Pharmacology, and Cannabis and Cannabinoid Research.

Preclinical and in-vitro studies

Cannabinoid receptor pharmacology

Published work with Dr. Yair Ben-Chaim at the Open University of Israel examined how cannabis terpenes act at the CB1 and CB2 receptors. It showed that selected terpenes activate these receptors and can increase the activity of THC, in some cases through synergistic effects, and that different terpenes behave differently. This is the mechanistic foundation for Bazelet's selected terpene enrichment, and it appears in two papers in Biochemical Pharmacology.

ASD neuronal models

A study with Prof. Shani Stern at the University of Haifa examined terpene-enriched CBD-dominant formulations in neurons derived from children with ASD, using patch-clamp recordings. Neurons from children with ASD showed reduced excitability and synaptic activity compared with controls; terpene-enriched CBD-dominant formulations improved both, while non-enriched formulations did not. Inflammatory conditions

A study with Prof. Timna Naftali at Sapir Medical Center examined cannabinoid formulations in cell models of inflammatory bowel disease and endometriosis.

Clinical trials

Bazelet's formulations are evaluated in randomized, double-blind, placebo-controlled trials with leading Israeli medical centers, in indications selected around the company's terpene-enrichment science. The program spans ongoing and completed trials in ADHD, autism spectrum disorder, and pain.

Terpene-enriched CBD oil in ADHD

A randomized, double-blind, placebo-controlled trial of a terpene-enriched CBD oil in children with attention-deficit/hyperactivity disorder, with Shamir Medical Center, Zerifin, headed by Prof. Matitiahu Berkovitch (Clinical Pharmacology and Toxicology Unit) and Dr. Orit Stolar. Designed for 120 children aged 6 to 18, naïve to cannabinoid treatment, comparing Bazelet's terpene-enriched CBD oil against a matched placebo, with inattention and hyperactivity assessed by standardized questionnaires and computer-based testing. A blinded phase is followed by an open-label extension. Recruitment ongoing. (NCT06542445.)

ADHD, long-term and target-dose evaluation

A continuation trial for children who completed the trial above, evaluating long-term use of the terpene-enriched CBD oil and the dose required for efficacy, comparing 5 and 10 mg/kg, with the same site and investigators and the same standardized measures. Recruitment ongoing.

Terpene-enriched CBD-dominant oil in ASD

A randomized, double-blind, placebo-controlled trial of a terpene-enriched CBD-dominant oil in children with autism spectrum disorder, with Shaare Zedek Medical Center, Jerusalem, headed by Prof. Adi Aran, head of the Neuropediatric Unit. Designed for 100 children aged 4 to 13, naïve to cannabinoid treatment, with behavioral problems and core ASD symptoms assessed by standardized questionnaires. A second phase compares the terpene-enriched CBD-dominant oil against a terpene-enriched, THC-free CBD oil to evaluate the contribution of THC. Recruitment ongoing. (NCT07199218.)

Completed

Cannabinoid formulation in acute radicular pain

A completed randomized, double-blind, placebo-controlled trial of a terpene-enriched THC:CBD (1:1) oil in 69 cannabis-naïve adults with severe acute radicular pain, with Hadassah Medical Center, headed by Prof. Elyad Davidson. Patients received a single high-dose formulation, low-dose formulation, or a matched placebo, with self-administered opioid use and pain intensity as primary outcomes over 24 hours. Primary results showed a substantial reduction in self-administered opioid use in the high-dose group compared with placebo, with no increase in pain scores. Manuscript under peer review. Terpene enrichment and THC in CBD-dominant oils (ASD pilot)

A completed randomized, double-blind, controlled crossover pilot in 12 children with ASD aged 4 to 10, with Shaare Zedek Medical Center, headed by Prof. Adi Aran, comparing a terpene-enriched CBD-dominant oil, a non-enriched CBD-dominant oil, and a terpene-enriched THC-free CBD oil, with behavioral and core ASD measures. Primary results indicated greater improvement on behavioral and core ASD measures with the terpene-enriched CBD-dominant oil than with the comparators. Manuscript under peer review.

Terpene-enriched CBD oil in treatment-resistant ASD symptoms

A published case report of an adolescent whose ASD-related symptoms, including severe aggression, were controlled with pure CBD in childhood but became unresponsive during puberty. Enriching the CBD with a selected blend of calming terpenes restored symptom control at a reduced CBD dose. Published in Frontiers in Pharmacology (2022).

60+ patent families since 2016

Since 2016 Bazelet has built a portfolio of 60+ patent families, with granted patents across multiple territories, covering its terpene-enriched formulations, its pre-milled products, and the production processes behind them. The portfolio is managed in-house and prosecuted with a leading US patent firm.

Bazelet's earliest filing in this portfolio, for terpene-enriched compositions, received an International Search Report finding all claims to have utility, novelty and an inventive step.
